Recipient's
blood is taken in two tubes : the first containing clear serum and the
second containing washed red cells. Two similar tubes are prepared for
the donor's blood. Two small tubes (reaction tubes) are marked as 'major' and 'minor'. In the tube marked 'major' add one drop of the recipient's serum. Add one drop of the donor's red cells (washed and suspended in saline to form 2 to 5% suspension). In the tube marked 'minor' add one drop of the donor's serum. Then add one drop of recipient's red cells (washed and suspended in saline to form 2 to 5% suspension. Mix well and incubate at RT for 90 minutes (or at 37 C for 30 minutes). Read for the presence of agglutinates by naked eye as well as under a microscope. |
