BASIC PREPARATIONS

Serum : Transfer from the plain container after identifying from the label and ensuring that the tube has the same identifying mark. Spin the tube to in the centrifuge obtain clear serum as supernatant.

Red Cell Suspension : Fill a large tube with Normal Saline (NS).
Add a drop of blood from unclotted specimen tube (Check both labels)
Mix well with pipette.
Spin the tube in the centrifuge.
Discard the supernatant saline and re-suspend the cells at the bottom of the tubes with fresh NS.
This is a suspension of "once washed" red blood cells RBC's
Repeat the procedure to get cells washed 3-4 times as required.
A five percent suspension has a colour similar to pink blotting papers.
